Product Description

Introduction
  • AutoPure Magnesium is a reagent kit for direct quantitative determination of magnesium in human serum and plasma on automated clinical chemistry analyzers.
  • AutoPure Magnesium is ready to use, single liquid reagent system.
  • With AutoPure Magnesium, the assay is linear upto 5 mg/dl (2.06 mmo1/1).
Principle

  • Magnesium ions reacts with xylidyl blue in an alkaline medium to form purple coloured complex.
  • Mg-+ Xylidyl blue -  Magnesium-xylidyl blue complex 
  • The intensity of the purple colour formed is directly proportional to the concentration of magnesium in the specimen and is measured photometrically.
  • Note: Calcium is excluded from the reaction by complexing with EGTA.

Shelf life

Stable till the expiry date indicated on the label, when stored at 2-8C.

On-Board Reagent Stability

  • 21 days at 2-8 C after opening . Protect the reagent from light and contamination.
  • Do not freeze the reagent.

Specimen Collection & Preservation

  • Collect sample using standard sampling tube. Although serum is  preferred , heparinized plasma can be used. Do not use EDTA plasma.
  • Haemolyzed samples should not be used. Magnesium in serum /plasma is stable for 7 days at 2- 8C and 1year at -20C. Centrifuge samples containing precipitate before performing assay. 

Calculations

Fully automated system automatically calculates the Magnesium concentration of each sample.

  • Results in mg/dl = Results in mmo1/1 x 2.43
  • Results in mmo1/1 = Results in mg/di x 0.412
  • Results in mmo1/1 = Results in mEq/I x 0.5
  • Results in mg/di = Results in mEq/I x 1.22

Calibration

For calibration, it is recommended to use C.f.a.s* or any other suitable calibrator material.

  • As per the requirement of quality control procedures.  

Quality Control

Each batch ofAutoPure Magnesium is assayed with atleast six quality control sera  Precinorm, Precipath, Biorad 1 Biorad II Accutrol Normal and Accutrol Abnormal prior to release.

To ensure adequate quality control, it is recommended that the laboratory should use a normal and abnormal commercial reference control serum. It should be realized that the use of quality control material checks both the reagent and instrument functions together.

If the control values fall outside the specified limits, each of the below criteria should be cross - checked and corrected:

  • Proper instrument function wavelength setting, light source and temperature control.
  • Cleanliness of probes and cuvettes.
  • Bacterial contamination of wash water used by the instrument.
  • Expiry date of the reagent kit.

Note:

Expected range varies from population to population. It is therefore recommended that each laboratory should establish its own normal range. For diagnostic purposes, the magnesium results should always be assessed in conjunction with the patient's medical history, clinical examinations and other findings.

Performance Characteristics Linearity

With AutoPure Magnesium, the assay is linear upto 5 mg/dl (2.06 mmo1/1). Determine samples with higher concentrations via the rerun function. On instruments without rerun function, manually dilute samples with higher concentrations using 0.9% NaCI or distilled/deionized water (e.g. 1 + 4). Multiply the result by the appropriate dilution factor (e.g. 5).

Interference

There is no significant interference in samples containing upto 60 mg/dl of bilirubin. Haemolysis interference is observed since magnesium is released from erythrocytes.

 Co - Relation Studies

A comparison of the magnesium determination using AutoPure Magnesium and reference reagent gave the following co-relation (mg/dl):

No. of samples measured : 62

The sample concentrations measured were between 1.5 and 5.2 mg/dl.
