INTRODUCTION
  • Infinite Liquid Microalbumin is a reagent set for quantitative determination of microalbumin in human urine.

PRINCIPLE

Infinite Microalbumin contains latex particles coated with specific anti-human albumin antibodies which react with microalbumin in the sample resulting in agglutination. Agglutination causes change in absorbance, measured at 540 nm (530 - 550 nm) & is proportional to the concentration of microalbumin in the sample.

PREPARATION OF WORKING SOLUTION

Swirl the latex vial gently before use and prepare the working solution freshly by mixing Reagent R1 and Reagent R2 in the ratio 4:1 as per the requirement. Microalbumin calibrator is ready-to-use.

REAGENT STORAGE, STABILITY & HANDLING

  • The kit components should be stored at 2-8 degree C and is stable till the expiry date indicated on the label. It is recommended to store the reagents at 2-8 degree C, tightly closed after use.
  • The working solution (4R1 + 1R2) is to be prepared fresh as per the requirement.
  • DO NOT FREEZE THE REAGENTS. Frozen Latex or Diluent could change the functionality of the test.
  • Contamination of the reagent should be strictly avoided.

COMPONENTS & CONCENTRATION OF WORKING SOLUTION 

  • R1 : Glycine buffer  80 mmol/L - 20m
  • R2 : Latex particles coated with anti-human albumin - 5 ml
  • R3:  Microalbumin Calibrator (Value mentioned on vial label) - 1 ml

SPECIMEN COLLECTION & PRESERVATION

  • 24-hour or random/first morning midstream urine is the specimen for this assay.
  • Specimen should be tested immediately, preferably within 12 hours of collection. Specimen can be stored up to 2 days at 2-8 degree C provided they are not contaminated.
  • Specimen should be free from particulate matter. Turbid or particulate urine
  • specimen must be clarified by centrifugation at 2000 rpm for 10 minutes.
  • Use the clear supernatant for testing. 

PROCEDURE

  • Reaction type: Fixed-Point
  • Reaction time: 10 mins. at 37 degree C/20 mins. at R.T. (25 -30 degree C)
  • Wavelength: 510 nm. (500 - 530 nm.)
  • Zero setting with: Reagent Blank
  • Blank absorbance limit: 0.300 Abs.
  • Sample volume : 0.01 ml (10 (If)
  • Reagent volume: 1.0 ml
  • Standard concentradivtion: 200 mg%
  • Linearity: 150 mg/L 

 PROCEDURE LIMITATIONS

  •  Linearity limit: If microalbumin concentration exceeds 150 mg/L, dilute the specimen with normal saline and repeat the assay. The result obtained should be then multiplied with the dilution factor to obtain correct microalbumin concentration.
  • Detection limit: Values less than 2 mg/L gives non-reproducible results.
  • Prozone effect: No prozone effect was detected up to 1000 mg/L.
  • Glucose (2 g/L), hemoglobin (10 g/L) and creatinine (3 g/L) do not interfere. Urea ( 1 g/L) and bilirubin ( 10 mg/dL) interfere with the | assay. 

QUALITY CONTROL

To ensure adequate quality control, it is recommended that each batch should include a normal and an abnormal commercial reference control serum. It should be realised that the use of quality control material checks both instrument and reagent functions together. Factors which might affect the performance of this test include proper instrument function, temperature control, cleanliness of glassware and accuracy of pipetting
